You can do both.
Editting layers has many options depending on the format of your data
and you should find plenty of information in the archives for both
editting shapefiles & postgis via mapscript.
Getting coordinates via a map click is just a matter of translating the
selected pixel coordinate into geographic coordinates based on the
extents of the map. The mapserver cgi does that automatically and there
are examples of how to do that in mapscript.
