How to create a layer at run time?

Can someone please tell me how to create a layer at run time?
(either in php or asp.net)
Also, how to plot a simple line on a layer, given coordinate points?

I have set up both php and .net sample applications and both of them are
working fine.

I am quite new to all this and any help from you is highly appreciated.
