According to my understanding

 

1 – It is possible to use file location as a url, or the file must be local
?

 

For an external file server, you can use alias. 

https://trac.osgeo.org/mapguide/wiki/ConfiguringAliases

 

 

2 – It is possible to use multiple files at once ? 

Put the multiple files in a directory if they représented a tiled map

 

3 – If using multiple files, which value to set the bounding box element in
config.xml ?

Each file should use its own word file.
