 YES!

 That was the Visual C++ Redistributable that was missing. I had the 32-bit
 version installed, but not the 64-bit. Both OSGeo4W Shell and QGIS Desktop
 are now running.

 I think this has to be marked as solved. But this is my first time with
 Trac and Tickets. How to proceed?
